Mass Spectrometry Analysis

Services All protein mass spectrometry-based analyses are performed using nano-flow high performance liquid chromatography (nHPLC) interfaced via electrospray ionization (ESI) to dual pressure linear ion trap mass spectrometers (LTQ Velos). All data analyses are conducted using Proteome Cluster. Reports consist of PDF summaries and MS Excel spreadsheets. Source data is available upon request, see Accessing your data.

 

Global Proteomic Profiling and the $1000 Proteome

The thousand dollar proteome is a global shotgun proteomic analysis of any sample type for $1,000. It includes a 6-step strong anion exchange fractionation and 12 hours of LC-MS/MS time on a state-of-the-art LTQ Velos, the fastest, most sensitive mass spectrometer available. This is a multidimensional proteomics identification technology (MuDPIT) assay as pioneered by the Yates Lab.

 

Protein Characterization

Proteins are digested prior to shotgun mass spectrometry on a state-of-the-art LTQ Velos tandem mass spectrometer (LC-MS/MS). Data is analyzed by up to four different search algorithms. Iterative searches are performed to maximize sequence coverage of identified proteins. This method is capable of identifying over 1,000 proteins in a complex mixture, however, sequence coverage will improve as the number of proteins decreases or enrichment of a set of proteins increases.
